Cells were inoculated into YPD (OD600=10⁻⁴), grown overnight, then diluted to OD600 0.01 in low iron media, incubated at 30°C for 5–6 hours, and harvested at OD600 0.5–0.6. Transcription factor gene deletion.

BPS: Cells were pre-cultured in liquid YPD (2% dextrose, 2% Bacto peptone, 1% yeast extract) rotating at 75 rpm at 30°C overnight. Cells were inoculated into 25 mL of pre-warmed (37°C) YPD + 500 µM BPS (bathophenanthrolinedisulfonate; iron chelator) at an initial OD600 of 0.2. Cultures were grown for 4 h with vigorous shaking (225 rpm) at 37°C, then harvested by vacuum filtration and immediately frozen at -80°C.
